Well stained thick film - thin but still good. Compare with other Plasmodium spp. (research) slides which have been prepared using different amounts of blood to determine the best method to diagnose the parasite infection.It is best to make the smears from native blood (ie. without anti-coagulant) as anticoagulant can distort the parasites. The thick film should be let dry for several hours-overnight before staining with Giemsa but if you need to stain within an hour, make the film a little thinner as here OR use Field's stain and treat carefully. If the leucocytes nuclei are dispersed and distorted, the film was not dry enough before staining and the parasites will also be distorted and possibly hard to recognise. It is important not to prolong the washing step as it will decolourise the parasites. As a gauge, the leucocytes? nuclei should be deep purple, a low pH or poor stain will also affect the quality. Identification should always be confirmed by examination of a thin film.
